✦ Synopsis

the authors want to correct the Methods section regarding the fixation/permeabilization protocols used for this study. The authors mentioned the use of 4% paraformaldehyde (PFA) on pages 2 and 3 to resuspend the cells lasting the final step before flow acquisition, when in reality 1% PFA was used. For the final wash of the BD-Foxp3 buffer, the authors used 2 ml of PBS instead of the 2 ml of permeabilization buffer described on page 3.

PBS was also used for all washes of the Caltag buffers.

The transcription factor FOXP3 plays a key role in CD4 + CD25 + regulatory T cell function and represents a specific marker for these cells. Despite its strong association with regulatory T cell function, in humans little is known about the frequency of CD4 + CD25 + cells that express FOXP3 protein

## Background: Cd4+cd25+ regulatory t cells (tr) can prevent or treat experimental murine colitis but little is known about their potential role in human inflammatory bowel disease (ibd). foxp3 is a transcription factor that plays a critical role in the development and function of cd4+cd25+ tr. the
