โฆ Synopsis
Electrospray tandem mass spectrometry is a useful tool for sequencing short (`20-mer) oligonucleotide chains. Product ion spectra obtained may lead to ion profiles which can be assigned to the different fragments generated from oligonucleotides. The technique can be applied not only to natural oligonucleotides, but also to modified oligonucleotides. This study demonstrates the use of tandem mass spectrometry to sequence a 7-mer oligonucleotide that had been adducted with the benzene metabolite, para-benzoquinone. The use of collision-induced dissociation of oligonucleotide ions of differing charge states enabled different fragmentation pathways to be observed, as did the use of differing collision energies in the collision cell. The carcinogen-adducted oligonucleotide was fully sequenced using these methods.
