Quantitative studies were made o n the effects of various additives such as procaine hydrochloride, dextrose, and citrate in reducing penicillin G procaine solubility in water. Aqueous vehicles were prepared i n which the solubility was lowered to a minimum of 0.036 mg./cc. at room temperature.
ONGER SHELF LIFE has been reported for L penicillin in aqueous procaine penicillin (penicillin G procaine, U. S. P.) suspensions containing various additives such as procaine hydrochloride and sodium citrate (1, 2). The increase in stability is attributed t o suppression of penicillin solubility. Morever, the rate of penicillin degradation in aqueous procaine penicillin suspensions appears to be associated primarily with the portion in solution. On this basis we devised a series of quantitative studies to determine the effects of various additives in suppressing penicillin solubility in such suspensions.
