β¦ Synopsis
Isoelectric focusing is a new method of separation in which amphoteric substances, in particular proteins, can be separated in a stable pH gradient owing to the differences in their isoelectric points. The method is extremely efficient and permits separation of proteins whose isoelectric points differ by only 0.02 pH unit. Further, the method enables the separated components to be characterized by a physicochemical constant, namely the isoelectric point. It is only recently that development of appropriate "carrier ampholytes" provided the prerequisites for practical use of the method. Several techniques of isoelectric focusing can be distinguished by the way in which the pH gradient is stabilized: 1. Focusing in a density gradient (e. g . saccharose); 2. focusing in polyacrylamide gel (either in flat gels or in an arrangement corresponding to disk electrophoresis); and 3. focusing in granulated gels (e. g. Sephadex or Bio-Gel).
