Two polyoma-TSTA-negative variants were selected independently from a polyoma fibrosarcomdMoloney lymphoma somatic hybrid, by repeated passages in polyoma-virus-preimmunized mice. One of the variants had lost all its polyoma DNA, while the other only retained a deleted piece of its integrated polyom

✦ Synopsis
Anfisera specifically directed against the TSTAs qf' adeno I 2 and polyoma nroitse tumors were absorbed with cells of other tumors induced by the same viruses. The antibodies adsorbed on tumor cells were subsequently eluted at p H 3. The eluate, absorbed serum and original serum were tested by the colony inhibition ( C I ) tcwhnique for coniplentent dependent cytotoxicity against adeno 12 and polyoma tumor cells syngeneic with the serum donors. Similar arlsorptioti-elrrtion experiments were performed with crude cell membrane fractions.
It was demonstrated that the anti-tumor antibody activity of the original serum can be absorbed away by both intact tumor cells and crude cell membrane fractions. The specific antibodies can subsequently be recovered by elution (at low p H ) jiom the cells used for absorption.

Antibodies against various H-2 antigens were separated by an adsorption-elution procedure. The findings might have implications for both antibody and antigen purifcation.
