The determination of L-ascorbic acid by utilizing lucigenin chemiluminescence with ascorbate oxidase was studied. The dehydroascorbic acid produced by the enzymatic reaction of L-ascorbic acid and ascorbate oxidase was found to be a more effective activator than L-ascorbic acid for the lucigenin chemiluminescence reaction. The optimum pH, temperature and incubation time for the enzymatic reaction were S.7,30Β°C and 10 min, respectively. The calibration graph was linear over the range 0.14-50 pg ml-'. The correlation coefficient for this line was 0.998. The detection limit (2~) was 0.1 pg ml-' and the relative standard deviation was 10% (n = 5) for a concentration of 0.14 pg ml-'. This method was applied to the determination of L-ascorbic acid in commercially available drugs and health drinks.
