β¦ Synopsis
A new one-step method for the preparation of polyacryhunide gels containing tritium and carbon-14 for liquid scintillation counting is presented. Forty-four hours after placing the gel slices in the scintillation fluid described in the text, the samples are ready for counting. This one-step method is compared to the method of D. Goodman and H. Martzura (1971, Anal. Biochem. 42, 481) and is shown not only to require fewer manipulations but also to eliminate the need for a heating oven and to reduce the reagent costs.

## Liquid scintillation counting of (3H) -and ('"C) -labeled compounds on solid supports is a common practice in biochemistry. Furlong has recently emphasized "local quenching" of tritium by filter paper, and has suggested that p's of higher energy might also be anomalously absorbed if they penetr
A method is described for analyzing the radioactivity of 3H-labeled RNA after separation by gel electrophoresis. The gels were soaked in 10% acetic acid and were sliced. The gel slices were dehydrated in alcohol, then saturated with toluene, and finally permeated with a toluene-based scintillation f
Electrophoresis in polyacrylamide gels is widely used for analysis and characterization of proteins and mixtures of proteins. It is frequently necessary to obtain estimates of the radioactivity of protein bands in the gels, and of the relative amounts of two radionuclides, generally 3H ,and l\*C. Ca
