✦ Synopsis


Most procedures for the isolation of DNA have been designed for the extraction of this material from calf thymus glands. Difficulties are, however, encountered when similar procedures are applied to isolate DNA from tissues such as rat and mouse liver, since the DNA is usually contaminated with RNA, part of which is resistant ta digestion with ribonuclease. Butler and Johns (1) reported that calf thymus DNA could be separated from added low molecular weight rat liver RN-4 by precipitation with lysine-rich calf thymus histone. We have found that this method can be used to purify preparations of liver DNA, since ribonuclease-resistant RNA is also detached from DNA. This appears to be a rapid and mild procedure which is especially useful when one wants to compare the i'n viva labeling of DNA and RNA.
