MATERIAL AND METHODS
Chemicals and cell culture
Retinoic acid and PMA were purchased from Sigma (St. Louis, MO). Retinoic acid was prepared weekly as a 10-mM stock solution in absolute ethanol and kept in the dark at -20°C. FBS was obtained from Hyclone (Logan, UT). Enzymes were purchased from Pharmacia/LKB (Montreal, Canada). Radiochemicals were from Amersham (Oakville, Canada) or Dupont New England Nuclear (Mississauga, Canada). The human neuroblastoma cell line SK-N-SH was obtained from the ATCC (HTB 11) and maintained in DMEM supplemented with 10% FBS, 2 mM glutamine and 35 pg/ml gentamycin. Media were changed twice a week, and cells were passaged on a weekly basis.
