A single-step procedure is small scale (annrox. 5mg) described for purifying a-lytic protease and its mutants on a from cell-free supematants of recombinant E. coli. Although free of non-enzyme pro&t, these preparations often contain fragments that arise from enzymatic self-digestion. Two strong cation exchange matrices are shown to vary greatly in their ability to separate such degradation products from intact enzyme.
