Simple radiochemical methods are described for the assay of acetyl-CoA synthetase, propionyl-CoA synthetase, and butyryl-CoA synthetase utilizing *%labeled acetate, propionate, and butyrate. Enzyme activity is followed by the measurement of in~o~ration of the l%Z-labeled short-chain fatty acids into their &A-derivatives.

Separation of the '*C-labeled reactant from its reaction product is achieved by rapid chromatography of the reaction mixture on Dowex anion exchange columns. A method is described to correct for inactivation of the enzymes by sucrose.
