โฆ Synopsis
A simple and highly sensitive method for quantitative amino acid and peptide analysis is described. It makes use of the reaction with [3H] dansyl chloride chromatography on polyamide plates, autoradiography at low temperature, and densitometry. This method was used for the quantitative analysis of the amino acids in a complete enzymic hydrolyzate of the synthetic hexapeptide H-Leu-Trp-Met-Arg-Phe-Ala-OH.
The limit of sensitivity for amino acids and small peptides is about 25 x lo-r5 mole with errors between 2 and 14% depending on the amino acid.
