✦ Synopsis


In 1963, Keston and Brandt

(1) first reported the determination of D-glucose anomers using a /3-n-glucose oxidase/peroxidase/chromogen system. Hill (2) developed an automated procedure using a similar system and measured deviations from equilibrium of anomers in the blood of dogs given epinephrine, glucagon, or insulin. In 1966, Jiirgensen and Jorgensen

(3) applied a similar determination of n-glucose anomers to the analysis of the reaction of glucosidases. However, these methods require more than 30 min for determination of each sample. The authors have devised a rapid polarographic microdetermination of n-glucose anomers using an oxygen electrode and P-n-glucose oxidase. The method consists of measuring oxygen consumption due to the oxidation of ,&n-glucose in fluids before and after attainment of mutarotational equilibrium. The advantage of this method is that the measurement requires less than 10 min for each sample. The procedure is described in detail here.
