✦ Synopsis

Abstract

The coupling of packed capillary liquid chromatography with electrospray ionization mass spectrometry has yielded an instrument capable of generating molecular weight information from very complex natural protein mixtures. The use of the packed capillary liquid chromatography columns offers two principle advantages over conventional liquid chromatography columns: high separation efficiencies and very low mobile phase flow rates (100 nL min^βˆ’1^). The low mobile phase flow rates permit the use of a relatively high ionic strength buffer (0.1% trifluoroacetic acid), While delivering a very low mass flux of the buffer to the mass spectrometer.

Surface layers (S-layers) are regularly ordered protein subunits found as the outermost cell envelope component of many bacteria. Most S-layers are composed of a single protein or glycoprotein species with a molecular weight varying between 40 and 200 kDa. Clostridium difficile is the most common ca

The sample amount is generally not restricted, however with LC‐ESI‐MS, a lack of sensitivity may still be observed with standard‐bore LC columns in isocratic mode. ## Abstract Identifying the Cys residues involved in disulfide linkages of peptides and proteins that contain complex disulfide bond patterns is a significant analytical challenge. This is especially true when the Cys residues involved in the disulfide bonds are closely spaced in the primary sequen
