✦ Synopsis

The oxygen demand of animal cells in microcarrier culture can be determined using a dissolved oxygen electrode. Continuous monitoring of the oxygen demand by human diploid fibroblasts shows it to be a good indicator of cell number. Furthermore, the oxygen demand of animal cells in large cultures can be supplied using membrane oxygenation using silicone rubber tubing. This method avoids the problems encountered with sparging the culture.

## Abstract The sampling method of polarographic analysis for dissolved oxygen in culture medium is unaffected by the presence of viable micro‐organisms. This method allows measurement of peak oxygen demand, critical oxygen concentration, and oxygen supply during actual mass propagation of an aerob

Both kLa and kL measurements were carried out by an unsteady state technique at impeller speeds ranging from 1.6 to 5.8 s-' in a mechanically agitated animal cell culture vessel of working volume l .5 L. Checks were made that the time constant of the oxygen electrode was negligible compared to the t
