✦ Synopsis
Mitochondrial D N A was isolated from mitochondrial fraction of the wheat germ [2]. Barn HI fragments of wheat m t D N A were cloned into pBR 322 plasmid. Screening of the bank was done by colony hybridization using 32p-labeled wheat mitochondrial tRNA as a probe [4]. Hybridizing Bam HI fragment of about 3 kb contained in the recombinant pBR 322 was partially digested with Sau 3a enzyme and subcloned in M13 vector [3]. For sequence analysis was selected one of the subclones that strongly hybridized with tRNA. The determined sequence is shown below:
