β¦ Synopsis
From a rRNA repeating unit of Sinapis alba (Brassicaceae) we have sequenced the 5.8S gene, the internal transcribed spacers [4] and the intergenic spacer with the 18S rRNA coding region [5]. Here we present the nucleotide sequence (3381 bp) of the 25S rRNA gene and thus complete the primary structure of the mustard ribosomal gene, which in total is 9005 bp long.
The nucleotide sequence of the 25 S rRNA gene is shown in Fig. 1 and was determined using both chemical and dideoxy sequencing methods [3,6]. The 25S coding region shows 97~ homology with the corresponding gene from Arabidopsis thaliana [8], which belongs to the same flowering plant family, and 93.5 ~o with Lycopersicon esculentum of Solanaceae [2]. The 18S coding region and the 5.8S gene show 98.8 ~o and 99.4~o homology respectively to A. thaliana [7,8]. The intergenic spacer of S. alba shows no significant homology to A. thaliana. An 84~o homology between S. alba and R. sativus of Brassicaceae [1] is found in the region from the downstream end of the repeating unit to the 18 S gene, which comprises more than 1 kb.
