✦ Synopsis


Using nuclear microscopy, extracellular diffuse amyloid deposits in fresh unstained brain tissue from AlzheimerΓ•s disease transgenic mice Tg2576 have been identified and analyzed for trace element content. Off-axis scanning transmission ion microscopy (STIM) images can be obtained which are similar to the images produced using direct STIM. Since the proton beam current required for off-axis STIM is compatible with PIXE and RBS, we can identify the plaque location and analyze for trace elements simultaneously. Analysis of the diffuse plaques showed an increase in the transition metals iron and zinc compared with the surrounding area of comparable areal density. This supports the theory that redox interactions between Ab and metals could be at the heart of a pathological feedback system wherein Ab amyloidosis and oxidative stress promote each other, possibly via Fenton chemistry.
