Dipeptidyl and tripeptidyl heterocycles designed to mimic peptide ketoamides and ketoacids were prepared and evaluated in vitro as inhibitors of human calpain I. Boc-Leu-Leu-imidazole (12) inhibited calpain I at low micromolar concentrations.
