The relatively insoluble drug, acetaminophen, administered as a solution and injected intraperitoneally in rats, has been shown to produce an antipyretic response. approximately twice that of the same dose administered as a suspension. This experiment indicates that EDw, LDw, and relative potency calculations may be inaccurately determined if dosage form influences on biological availability of insoluble drugs are not considered in the design and evaluation of animal drug testing procedures.
