A new method of hydrolyzing proteins and peptides without racemizing the amino acids has been developed. This method consists of performing a brief partial chemical hydrolysis for \(15 \mathrm{~min}\) in \(6 \mathrm{M} \mathrm{HCl}\) at \(80-90^{\circ} \mathrm{C}\), followed by an enzymatic hydrolys

I. I n t r o d u c t i o n
I n continuation of the work published in 1967 (IWAMURA et al.), planktonic organisms cultured in the laboratory (Chlorelb and Phaeodactylum) as well as those collected from natural waters were analyzed for their contents of chlorophyll, protein, RNA and DNA in the hope of establishing new methods of measuring planktonic populations in natural waters. Some improvements were made in the assay techniques, but, despite our efforts, the procedures could not l) This work was supported b y a part of the Special Project Research Fund (for "Studies on the dynamic status of biosphere") granted from the Ministry of Education to our research group, headed by Prof. HIROSHI TAMIYA. 9.

A method to determine the relative rates of stable and unstable RNA synthesis is presented. The method employs the antibiotic rifampicin. For exponential phase Escherichia coli B/r growing at rates between 0.67 and 2.10 doublings per hr (3.2-fold range in growth rate), the fraction of the instantane
Fourier transform (FT)-IR spectroscopy was used to characterize and check for contamination in lignothioglycolic acid (LTGA) preparations from radish seedlings. LTGA, prepared by the conventional technique of extracting plant material first with 50% (v/v) aqueous methanol, was so highly contaminated
The spectrophotometric periodate-resorcinol method for the measurement of free, bound and total N-acetylneuraminic acid has been modified. The proposed method improves sensitivity by 27%, reduces the time of analysis by 33-52% and addresses interferences from biological substances. The improved proc
Two standard titrimetric procedures for determination of free fatty acids in vegetable oils and in oils extracted from oilseeds were examined using rapeseed oil, illipe nuts and fat, and groundnuts and groundnut oil as test materials. Both procedures involve dissolution of the fat in an organic solv
