Conditions for mat,ing of Sacchnromycopsis lipolyfica cells were investigated to cnhance zygote format,ion. Conjugat,ion frequencies could be increased about twenty five fold by rising ce11s of tho late logarithmic growt)h phase and a yeast'-extract-malt medium t'o induce ma'ting. A procedure based o n thcsr: results is descrihcd and has been used to carry out complrmentat.ion anitlysis in 8. Zipolyticn much more rapidly.
