✦ Synopsis

The presence of prostaglandin E (PGE) in rheumatoid synovium was investigated by immunoelectron microscopy; horseradish peroxidase-conjugated antibodies were used to stain the PGE. PGE was stained mainly in cytoplasmic granules of macrophage-like cells of the rheumatoid synovium. These granules had the appearance of lysosomes. The findings suggest that the cell that produces PGE in rheumatoid synovium is mainly a cell of the macrophage lineage.

Increased production of prostaglandins, especially prostaglandin E (PGE), has been described in rheumatoid joints (1-3). Substantial quantities of PGE have been detected in rheumatoid synovial fluids and rheumatoid synovial cell culture media. Furthermore, strong staining for PGE has been demonstrated in rheumatoid synovial tissue by indirect immunofluorescence (4). The type of prostaglandin produced by rheumatoid synovium is mainly PGE2 with lesser From the Rheumatic Diseases Unit,

Objective. Microsomal prostaglandin E synthase 1 (mPGES-1) catalyzes the formation of PGE 2 from cyclooxygenase-derived PGH 2 . Microsomal PGES-1 is induced by proinflammatory cytokines and is strongly linked to conditions that result in high PGE 2 biosynthesis. PGE 2 contributes to the pathogenesis
