Spironolactone (I), a potassium sparing diuretic, is widely used in the treatment of edema. Metabolic degradation starts with rapid deacetylation, leading to the short lived a-thiol [l]. Subsequent elimination of hydrogen sulfide yields canrenone (II), which exists in enzymatic equilibrium with canrenoic acid (Ill), which is the predominating form at pH values higher than 8 [2]. Under physiological pH conditions canrenone is relatively stable. I1 111 In the past canrenone has been determined almost exclusively by fluorimetry [3,5,6]. Reaction with 65% sulfuric acid generates a fluorophore with a trienone structure. Since this structure is produced not only by canrenone but also by other metabolites, these assay methods lack specificity. Some authors [7,8,9] describeGLC methods-also in combination with MS detection-for determination of canrenone in biological samples.
