## Abstract An explicit relation between the percentage (__G__ + __C__)‐content __X__~__GC__~, and the melting temperature (__T__~__m__~) of a natural DNA, is derived froma a statistical mechanics calculation with due consideration given to the base pair bonding and stacking interactions. A rapid high performance liquid chromatographic (HPLC) method is described which offers a fast quantitative procedure for determining the percentage of guanine (G) plus cytosine (C) in DNA preparations after acid hydrolysis in 88% formic acid. As little as 10 Fg of DNA is amenable to this analysis.
