The preparation and regeneration of protoplasts from Gibberella .fujikuroi was examined. High yields of protoplasts were obtained by using a mixture of lyophilized enzyme preparation from the snail Helix pomaria and a laboratory prepared mycolytic enzyme system from Trichodernia harzianurn. The physiological stage of the mycelium and the mycelium amount were found to be important factors in the determination of the protoplast yield. Regeneration could be achieved on the surface of culture media osmotically stabilized with NaCI. KCI, mannitol or sorbitol.
