By considering a DNA molecule as a random sequence of four letters, a mathematical model of two important steps in DNA cloning processes is described: degradation of large DNA molecules using restriction enzymes, and replication of the resulting DNA fragments by viral vectors. The model is used to evaluate the feasibility of obtaining target gene loci using a large number of restriction enzymes, and to estimate the fraction of genomic DNA that is clonable.
