Triazolyl derivative of the thymine mononucleotide (the compounds are dichlorophenyl phosphates). quasi-molecular ion m/z 818 associated with the base fragment m/z 176, the respective underivatized peaks being m/z 867 and m/z 176. The atomic composition of the modified base has been directly determined by precise measurement of the ratio of the masses of these two bases anions. The mixture of the products was condensed with thymine in order to obtain the dimer 2, I(MeO),Trl dTdT. The triazolyl adduct of the dimer has a quasi-molecular ion peak at m/z = 1198 (normal value at m/z = 1147). This example shows how it is possible to monitor by FAB mass spectrometry the condensation step in oligonucleotide synthesis. J.
