β¦ Synopsis
On the basis of reports that some calcium channel blockers impair the elimination of some drugs, the effect of nifedipine on the disposition of antipyrine and theophylline was assessed in healthy volunteers. Antipyrine half-life of 10.04 f 1.43 h (mean f SD) after a week intake of nifedipine (20 mg twice daily) was not significantly different from the control value of 10.64 f 2.15 h; nor was that of 10.02 & 1.49 h after 2 weeks pretreatment with the calcium channel blocker in eight healthy volunteers. Control antipyrine clearance (ml min-I) of 4440 10.58 was not significantly different from that of 45.66 f 9.34 and 46.87 f 9.63 after nifedipine pretreatment of 1 and 2 weeks, respectively. Similarly volume of distribution was unaltered: 0.601 f 0.074, 0,591 f 0.078 and 0.602 + 0.051 1 kg-', respectively.
A week pretreatment with nifedipine did not significantly alter either of theophylline half-life (7.32 f 0.81 h (control) to 7.50 f 0.80 h) or clearance (42.10 f 5%4mlmin-' (control) to 43.77 f 4.00 ml min-I) in six volunteers. However the change in volume of distribution: 0.451 f 0.053 1 kg-l (control) to 0.483 f 0.062 1 kg-' was significant (p < 0.025). Generally, theophylline plasma levels were lower after nifedipine pretreatment and the difference was significant at 2 and 4 h post-dosing @ < 0-05).
It is suggested that nifedipine, unlike diltiazem and verapamil, is unlikely to interfere with the functional integrity of the hepatic mixed-function oxygenase enzymes, but might displace theophylline from plasma protein.

The effect of domperidone (2 mg kg-') on the pharmacokinetics of a single oral dose of theophylline (25 mg kg-') was studied in the rat. Theophylline concentrations were measured serially for 12 h using an HPLC technique. Domperidone did not have any significant effect on any of the four parameters
The efSect of early nifedipine therapy on acute renal allograft rejection was studied in I70 adult cadaveric transplant recipients. Acute rejection occurring in the jirst 3 months after transplantation was diagnosed by Tru-cut biopsy and the severity of each rejection episode assessed histologically
## Abstract Theophyllineβ7βacetic acid (T7AA) was administered intravenously to rabbits so that its distribution and rate of elimination could be ascertained. The analytical techniques used were liquid scintillation counting (^14^C) and high pressure liquid chromatography. T7AA was eliminated rapid
The disposition of theophylline was studied on four occasions in eight healthy adult males. The control mean theophylline half-life and clearance were 7.32 h and 086 ml min-Ikg-', respectively. After 5 days pretreatment with placebo the corresponding values of 7.01 and 0-88 were not significantly di
