✦ Synopsis

A fast and reliable enzymic method is presented for differential quantitative determination of acetyl-CoA and acetyl-dephospho-Cot either present in mixtures or separately. The sum of both compounds is determined by the assay with citrate synthase (EC 4.1.3.7), as known. Conditions are given under which acetyl-CoA but not acetyl-dephospho-CoA, present in the same solution, reacts with glyoxylate on malate synthase (EC 4.1.3.2). Thus, acetyl-CoA and acetyldephospho-CoA are easily determined quantitatively by use of the two enzymic assays.

The rate of acetylcholine (ACh) synthesis was found to depend on the activity of choline acetyltransferase (ChAT) and on the concentrations of the two substrates of this enzyme, choline and acetyl-CoA. We present gas chromatographic-mass spectrometric assays for (i) the concentration of sulfamethoxazole and (ii) the concentration and molar percentage enrichment of acetyl-sulfamethoxazole in biological fluids. The compounds are extracted with ethyl acetate, derivatized with either diazomethane or p
