Polycyclic aromatic hydrocarbons ( (\mathrm{PAHs}) ) in oyster tissues were isolated by saponification with aqueous potassium hydroxide and hexane partitioning. Extracts were purified by adsorption chromatography using silicic acid and alumina oxide followed by gel chromatography with either Sephadex LH-20 or Bio-Beads S.X8. Measurement of PAHs were performed by high-performance liquid chromatography (HPLC) with ultraviolet absorbance and fluorescence detection. Recovery trials using NBS 1647a-spiked oyster homongenates revealed that the use of adsorption chromatography followed by gel chromatography was not effective in purifying extracts to allow the determination of lower ring PAH by HPLC or higher ring PAH by gas chromatography-mass spectrometry.
