Gas-liquid chromatographic procedures for the determination of 4-aminopyridine in human and animal plasma are reported. The procedures involve the addition of an internal standard 3-methyl-4-aminopyridine to plasma followed by extraction into methylene chloride/isopropranol under alkaline conditions. The isolated amines are converted to their pentafluoropropionyl or monochlorodifluoracetyl derivatives and are quantified after chromatographic separation using either electron-capture or nitrogen-phosphorus detectors. The sensitivity of these procedures is such that nanogram amounts of 4-aminopyridine can be readily determined.
