N,O-bis(trimethylsilyl)trifluoroacetamide, N-methyl-N-(tert-butyldimethylsilyl)trifluoroacetamide and trifluoroacetic anhydride have been investigated as derivatizing agents for GC/MS determination of N 3 -methyl-5'-deoxy-5-fluorouridine (N 3 -Me-5'-dFUR), a metabolic product of the anticancer pro-drug doxifluridine. Mass spectral information is given and some fragmentation patterns are discussed. Relative merits of the three derivatizing agents are also discussed. Examples of applications to spiked serum and urine samples are reported. Detection limits of the order of 10 nM were obtained when operating in selected ion recording mode.
