โฆ Synopsis
The effect of metoprolol on adriamycin-induced cardiotoxicity (AIC) was investigated. Adriamycin was administered to rats i.v. as a single dose of 6mg/kg, and metoprolol was continuously administered by an implanted osmotic pump (1 umol/hr) during four weeks.Ca 2+ -ATPase and beta-adrenoceptor density and affinity were determined.The Ca g+-ATPase activity was shown to increase ( 45 + 3.8 vs 23 + 1.2 umol Pi /mg/hr,P<0.01 ) in AIC compared to controls.After treatment with metoprolol the Ca2+-ATPase activity was found to decrease (29 + 2.1vs 45 + 3.8 umol Pi/mg/hr,P<0.01) in AIC. In contrast,no alteration in betaadrenoceptor density was observed, neither did the high-affinity agonist binding to beta-adrenoceptor change in AIC with or without metoprolol treatment.These results suggest that metoprolol prevents Ca 2+overloading in AIC possibly via a beta 3-adrenoceptor mechanism.
