✦ Synopsis

The molecular organization of the microtubules in a cilium or a sperm flagellum is not resolved in preparations fixed by the standard fixatives, osmium tetroxide or gh~.taraJdehyde followed by osmium tetroxide. The protofilaments become visible only after the addition of tannic acid to the glutaraldehyde, as was first demonstrated by Mizuhira. He and his successors have used osmium tetroxide as a post-fixative, in spite of its harmful action on many parts of the cytoskeleton. In an attempt to improve the fixation quality of cilia and flagella we have tried Mizuhira's technique modified by omi.tting osmium post-fixation.
