✦ Synopsis
Two procedures are described for column chromatographic separation of the (I! and p subunits of brain tubulin using hydroxylapatite in the presence of(i) urea or (ii) sodium dodecyl sulfate and /3-mercaptoethanol. In the first system the a and /3 chains are partially resolved, and in the second the subunits are resolved into three peaks which we designate a,, (Ye. and p.
METHODS AND RESULTS
Preparation of Tub&in
Tubulin was prepared by the method of Weisenberg et al. ( 8) with some variations. Fresh calf brains were obtained from a local slaughterhouse. The cortexes of the brains were cleaned and, in a typical experiment, 800 g were homogenized in 800 ml of buffer (10 mM sodium phos-243 Copyright
