trans-3-Hydroxy-L-proline is frequently eluted from amino acid analyzer columns as a double or asymmetric peak. This behavior was shown to depend on the size of the hydroxyproline sample, the pH, and the nature of the eluting buffer. Comparison of a number of organic acids used as eluting buffers suggested that the hydroxyl group of malic or citric acid participates in reversible complex formation with 3-hydroxyproline. Evidence for such chemical interaction was obtained by NMR spectrometry.

' It is of interest that apparent chromatographic heterogeneity of trcrrrs-3-methoxy-DLproline was explicitly noted in this paper (9).
