Synthesis of Elenic Acid, an Inhibitor of Topoisomerase II.

-Stereospecific synthesis of title compound (VII) is achieved in five steps and an overall yield of 26% from iodophenol (I) and eicosyne (II), involving novel zipper reaction of alkyne (III) and one-pot elaboration of the terminal alkyne in (IV) to an (E)-Ξ²,Ξ³-unsaturated ester containing an Ξ±-stereocenter. This method is a general short, efficient route for the preparation of structural analogues.
