Efficient Large-Scale Synthesis of 4-Phenyl-3-butyn-2-one, a Key Intermediate for a Novel Potent Adenosine Antagonist.

-A practical and facile large-scale synthesis of the 4-phenyl-3-butyn-2-one (IIIa) is presented based on the reaction of the in situ formed acetylenic Grignard reagent with two equivalents of acetic anhydride (II). The amount of acetic anhydride plays a crucial role on the reaction outcome since excess of the in situ formed acetic acid causes side reactions. The method can be readily extended to the acylation of several alkynyl derivatives (6 examples). -(ZANKA, A.;
