A procedure is described for the rapid preparation of a partial cyanogen bromide digest of horse heart cytochrome c. This digest serves as a calibration mixture for estimation of pep tide molecular weight in the range 500 to 20,000 by exclusion chromatography in denaturing solvents. The elution profile of the calibration mixture from Sephadex G-50 equilibrated and developed with 10% formic acid is described by the relationship log M = 4.79 -0.42
