Acute promyelocytic leukemia (APL) is usually associated with the translocation t( 15; I7)(q22;q 12-2 I), which disrupts the retinoic acid receptor alpha (RAM) gene on chromosome I7 and the PML gene on chromosome 15. We report a patient with typical APL without the common t( 15; 17). Cytogenetic stu

β¦ Synopsis
D N A studies of the translocation t( 15; 17) in acute promyelocytic leukemia (APL) have shown that the retinoic acid receptor alpha (RARA) gene on chromosorne I7 is juxtaposed to the promyelocytic leukemia (PML) gene on chromosome 15. The PML breakpoints have been mapped to1 3 clusters: bcr I, bcr2, and bcr3. W e have examined the PML breakpoint distribution in a series of 33 Chinese patients with APL. Twenty-two patients fell within bcr I, 2 within bcr2, and 9 within bcr3. The primary structure of the reciprocal chromosome translocation joints of one patient and that of their normal counterparts have been determined and compared to those of 2 previiously reported cases. These studies revealed possible topoisomerase II cleavage sites close to the breakpoints and suggested implications of D N A attachment sites to nuclear matrix. W e propose that these features are relevant to the process of illegitimate recombination generating the translocation. Genes Chrom Concer 6: 133-139 ( / 993).

Molecular analysis of the t ( 15; 17) translocation in 70 patients with acute promyelocytic leukemia (APL) confirmed that the breakpoints of chromosome I5 were located in two regions of the promyelocytic leukemia (PML) gene, mainly introns 3 and 6, whereas the breakpoints of chromosome I 7 were cons
## Abstract We compared genomic breakpoints at the __PML__ and __RARA__ loci in 23 patients with therapyβrelated acute promyelocytic leukemia (tβAPL) and 25 de novo APL cases.Eighteen of 23 tβAPL cases received the topoisomerase II poison mitoxantrone for their primary disorder. DNA breaks were clu
## Abstract ## BACKGROUND The clinical relevance of mutations of the __FMS__βlike tyrosine kinase 3 (__FLT3__) gene in specific cytogenetic subgroups is not clear. The authors examined internal tandem duplication (ITD) and Asp835 mutations of __FLT3__ in patients with acute promyelocytic leukemia
The nucleoporin gene NUP98 has been reported to be fused to 9 partner genes in hematologic malignancies with 11p15 translocations. The NUP98-HOXA9 fusion gene has been identified in acute myeloid leukemia (AML) and chronic myelogenous leukemia with t(7;11)(p15;p15). We report here a novel NUP98 part
