✦ Synopsis


Gout is a disease i n which deposition of uric acid plays an important role. Many of the factors that affect urate solubility have been extensively investigated (l), but the solubility is essentially a function of urate ion concentration, or more precisely of urate activity. If urate, like many other anionic substances, is bound to human serum albumin, its activity will be decreased, thus modifying its physiologic activity. I t therefore became important to determine the extent, if any, of binding of urate ion to human serum albumin under physiologic conditions of pH 7.4, ionic strength 0.16, and a temperature of 38OC. T h e human serum albumin-urate interaction was investigated by a continuous ultrafiltration technique (2) that permitted precise measurement of binding over the range of urate concentrations encountered clinically. This procedure allowed the determination by Scatchard analysis of the binding constants and the number of binding sites.
