The rate of DNA synthesis in HeLa S3 cells, as measured by incorporation of C14-labeled thymidine, is strongly dependent on protein synthesis at all times during the S phase. The relation between the rate of DNA synthesis and the rate of protein synthesis is linear when measured two or three hours a

β¦ Synopsis
A 17kD heparinβbinding protein (HBp17) has a biphasic doseβdependent effect on DNA synthesis in 3T3 cells. Maximal stimulation of DNA synthesis occurs at 8ng/ml HBp17, but a halfβmaximal inhibition occurs at βΌ500ng/ml. This inhibition can easily be reversed by addition of 400pg/ml aFGF or 100pg/ml bFGF, whereas EGF had no effect. This biphasic action of HBp17 was also seen in human umbilical vein endothelial cells (HUVEC), whereas it was not found in the malignant cell line, A431βAJC. The functional relationship between HBp17 and FGF is discussed.

## Abstract Cellβfree protein synthesis is a useful research tool and now stands poised to compete with in vivo expression for commercial production of proteins. However, both the extract preparation and protein synthesis procedures must be scaled up. A key challenge is producing the required amoun
