With the exception of a calorimetric method (1)) now known to give excessively high results, the assay of tryptophan in urine has been possible only by microbiological methods. At,tempts to apply the spectrofluorometric method of Duggan and Udenfriend (2) directly to urine or to partially partitioned samples using Dowex 50 with alkaline elution were unsuccessful2 The method of quantitative paper electrophoresis described below was successfully applied to urine. In the course of a comparative study of the electrophoretic method with the microbiological assay a large difference between the apparent tryptophan content of urine of normal and phenylketonuric individuals was observed by the microbiological method but not by the electrophoretic method.
