Extraction with ethyl acetate followed by gas-liquid chromatography on the extract provides a useful analytical method for phenol oxidase studies where it is desired to monitor directly the concentrations of volatile phenolic and catecholic substrates. A packed column with 10% DC-710 on Chromosorb W-AW-DMCS is recommended for such compounds as p-cresol, catechol, and 4-methylcatechol. p-Methoxyphenol is a suitable internal standard to minimize problems in the handling and injection of the extract. Examples of cresolase and catecholase kinetic runs are presented.
