Mutations affecting the glycogen biosynthesis in E. coli can be mapped at three different loci, glg A, glg B and glg C lying between asd and mal A. Transduction tests suggest the following order for the genes in this region: mal A--glg A--glg C--glg B--asd.

The promoter of the threonine operon was joined to the structural genes of the lac operon in Escherichia coli K 12. The synthesis of beta-galactosidase was thus repressed by threonine plus isoleucine in the fusion strains. To isolate mutations which affect the expression of the threonine operon, alt
The dam gene of E. coli can be inactivated by insertion of Tn9 or Mud phage. Strains bearing these mutations are viable indicating that the dam gene product is dispensable.
We isolated strains of Escherichia coli K 12 in which the lac structural genes were fused to the structural genes of the guaBA operon. These strains were used to isolate regulatory mutations that increased the expression of the guaBA operon under normal repressing conditions as compared to the wild
