A noncommercial Hadamard transform microscopic image analysis system was used to measure fluorescence emitted by single cells. A method for subtracting the native fluorescence of the Zephyranthes candida(Lindl.) Herb. pollen cell was developed based on its correlation to pollen area. Acridine orange and ethidium bromide were used as fluorescence probes for pollen DNA quantitative analysis, and the results obtained by the two methods were compared.
