2-Mercaptoethanesulfonic acid (coenzyme M, HS-CoM), methylcoenzyme M (CHa-S-CoM), acetylcoenzyme M (CH,CO-S-CoM), 2,2'-dithiodiethanesulfonic acid [(S-CoM),], and bromoethanesulfonic acid can be simultaneously and conveniently determined by isotachophoretic analysis. Amounts as low as 10 pmol can be detected. The reproducibility of the method is within 2%. The reduction of (S-CoM)% and the formation of CH,-S-CoM from HS-CoM and methanol by dialyzed cell-free extracts of Methanosarcina barkeri were studied.
