✦ Synopsis


An activity stain for the detection of L-serine dehydratase in gels is described. After electrophoresis, the gel is incubated with L-serine, and the pyruvate formed is detected by its reaction with 0.6% 1,2-diamino4nitrobenzene in 2 M WC1 at 90Β°C. A red band on a yellow background indicates the position of L-serine dehydratase.

Staining of polyacrylamide gels with methylene blue prior to silver staining increases band resolution and sensitivity. This method permits resolution of multiple bands less than 1 mm apart, and is able to detect bands containing only 100 pg of RNA.
