โฆ Synopsis
The high solubility of cyanogen bromide in N-methyl-2-pyrrolidone has afforded an efficient means of controlling the amount of the reagent used to activate an agarose preparation. By controlling the activation step, one can determine the final concentration of the leash moiety. The upper limits of ~-aminocaproic acid incorporation seem to be about 5, 20, and 60 tzequiv/ml gel, respectively, for 2, 4, and 6% agarose gels. It is important to conduct the cyanogen bromide reaction at 4-10ยฐC, but the coupling step may be done at room temperature.
